OPM-293 Platform is a combination of chemically-defined media and feeds, specially designed and developed for high-efficiency transient transfection and protein expression of HEK293 cell lines, as well as for high-efficiency proliferation of Adeno-associ_x0002_ated virus (AAV), Lentivirus (LV) and Adenovirus (AdV). OPM-293 Platform perfectly meets the needs of protein expression, virus production, cell therapy and gene therapy.

OPM-CD Trans293 cell culture medium supports high level transient expression of Lentivirus (LV), Adenovirus (AdV) and Adeno-Associated Virus (AAV).

OPM-CD Trans293+293 ProFeed cell culture medium supports high level transient expression of Lentivirus in conjunction with 293 ProFeed. The LV titer can increase more than 50%, reaching 1x10⁷ TU/ml

OPM-CD Trans293 cell culture medium supports the highest level of transient expression of AAV compared to some other commercial media. Prior to process optimization, the AAV titer can already reach 2.29x10¹⁰ vg/ml

OPM-CD Trans293 cell culture medium supports comparable level of transient expression of Adenovirus (2x10⁹ IFU/ml)

FAQ

What level of protein titer can I expect after transient transfection?
- Usually, antibody titer can reach 50 - 300mg/L
After protein expression in HEK293 cells using OPM 293 CD 05 medium,nickel containing chromatography column for downstream purification often seem to lose nickel.How can I improve the purity of the final product?
- We recommend using chromatography column without Nickel, e.g. Ni Excel from Cytiva.
Are OPM 293 cell culture media suitable for cell & gene therapies?
- OPM 293-CD05 with feed 293-Profeed can culture 293FT cells to produce lentivirus (LV), a vector for lentivirus for cell therapy.
What do I do if OPM 293-Profeed leads to declining of cell viability after transient transfection of HEK293 cells?
- We recommend to explore the optimal concentration of 293-Profeed. As a guideline, please try to add 5%, 3%, and 7% respectively.
What is the range of acceptable endotoxin level in OPM cell culture media?
- (1) Endotoxin level in basal media is controlled at ＜ 1.0 EU/ml, (2) Endotoxin level in feeds is controlled at ＜ 10 EU/ml, (3) Endotoxin level in ultra-concentrated feed is hard to measure because high PH can disturb the detection. For such concentrated feeds we usually perform internal quality control. However, our customer can request for including the analysis in the CoA, such as we did for the supplement CDFS08.
What is OPM's definition of chemically defined (CD) media? What about media containing Insulin and IGF-1?
- As per globally accepted definition, media containing insulin and IGF-1 are considered as CD, but if these ingredients are included in the media, we will clearly communicate as such to the customer.
Do OPM's feeds contain glutamine?
- None of our current catalog feeds contains glutamine. For specific products containing glutamine, we will indicate them.
